Dengue, a flavivirus, is transmitted by Aedes aegypti and Aedes albopictus mosquitoes. Symptoms typically manifest three to fourteen days post-infection. The dengue NS1 test, which stands for Dengue non-structural protein 1, is utilized for early dengue fever diagnosis by detecting the presence of dengue NS1 antigen in the blood. This test allows rapid detection on the first day of fever, preceding the appearance of antibodies by 5 or more days. In primary Dengue infections, IgM antibodies become detectable 3 to 5 days after the onset of fever and usually persist for 30 to 90 days. In endemic regions, most Dengue patients experience secondary infections, leading to elevated levels of specific IgG antibodies, either before or concurrently with the IgM response.

Principle
Dengue NS1 : The Rapid Dengue NS1 Antigen test involves adding a blood sample to the device, where it reacts with gold-conjugated Dengue NS1 antibodies. This forms a complex that moves onto a nitrocellulose membrane, creating a colored Test Line if Dengue NS1 antigens are present. The Control Line, generated by the reaction with goat anti-rabbit IgG, serves as a procedural control, indicating a valid test when both lines appear.
Dengue IgG/IgM : The Dengue IgG/IgM Antibody test employs a lateral flow chromatographic immunoassay. When the sample containing Dengue-specific IgM and/or IgG antibodies is added, it reacts with colloidal gold particles carrying Dengue-specific recombinant antigens, forming a complex. This complex then produces colored bands on the test and control lines, with the appearance of both indicating a positive result, while the control line alone signifies a negative result, ensuring the test's procedural and reagent integrity.

Limitation
- The test is for in vitro diagnostic use only.
- This test detects the presence of Dengue NS1 antigen in the specimen and should not be used as the sole criteria for the diagnosis of Dengue virus infection.
- Secondary infection may show low IgM and high broadly reactive IgG; cross-reaction possible.
- Negative result may occur in short-term or second-time infections; retesting recommended if suspicion persists.
- Diagnosis and treatment by a physician based on comprehensive clinical and lab evaluation.
- Antibody presence doesn't determine treatment success.
- Optimal dengue testing time is 6-14 days after fever onset.
